How should the 3-marker wand be held? Should the markers be held vertical or horizontal, or should one spin the wand throughout the calibration?

Any and all ways is the best way to do it! I generally wand with the orientation changing either fast or slow, and usually finish by going through the volume rapidly spinning the wand. The more samples and the more positions those samples are in, the better your calibration will be!
