Where are the camera setup stored? I mean, they are stored in the calibration file, and loaded in the camera when you load that file, or they are stored directly in the camera when I adjusted in the Preview mode of the calibration tool?

The camera setup is stored in the calibration file. Once you've loaded a calibration file into the point cloud library you can read out the resulting camera positions and orientations as well as the values you've used for the point cloud parameters, etc.
